介紹
KRAS 突變?cè)趲追N癌癥中都有發(fā)現(xiàn),例如結(jié)直腸癌、肺癌、甲狀腺癌、胰腺癌、膽管癌。KRAS 突變通常出現(xiàn)在外顯子 2 的密碼子 12 和 13 上,導(dǎo)致 p21-ras 蛋白的生長(zhǎng)信號(hào)異常變化。這種與細(xì)胞生長(zhǎng)和分裂有關(guān)的異常變化可能通過(guò)傳遞過(guò)量信號(hào)引發(fā)癌癥發(fā)展過(guò)程。KRAS 突變的存在通常與藥物反應(yīng)的預(yù)后標(biāo)志相關(guān)。例如,KRAS 突變被認(rèn)為是酪氨酸激酶抑制劑(如吉非替尼(Iressa)或厄洛替尼(Tarceva))藥物反應(yīng)的強(qiáng)預(yù)后標(biāo)志。最近,KRAS 突變經(jīng)常在結(jié)直腸癌中檢測(cè)到,可能與用于結(jié)腸癌治療的西妥昔單抗(Erbitux)或帕尼單抗(Vectibix)的藥物反應(yīng)有關(guān)。因此,檢測(cè) KRAS 突變對(duì)于確定結(jié)直腸癌或肺癌患者的藥物耐藥性,并有助于癌癥治療。
